I found a family tree at Ancestry.com which states that John W. Clanton was married to Mary Elizabeth Hughston, younger sister of Sanford and William. They were married 4 Jan 1849 in Lincoln County, Tennessee.

I located them on the 1850 census and the 1860 census of Lincoln County, Tennessee.

John W. Clanton and his wife, Elizabeth, are buried in Bethesda Cemetery in Senatobia, Mississippi.
